چکیده

Sodium-dependent calcium efflux from rat liver mitochondria has been studied as a function of mitochondrial calcium loads (2 to 40 nmol/mg) and extramitochondrial sodium concentrations (5 to 40 mM). The resulting data can be fit to a terreactant model which exhibits simultaneous kinetics (Le. both sodium and calcium must be bound simultaneously for transport to occur). The Hill coefficients for the calcium and sodium dependences were 1.0 & 0.1 and 2.0 & 0.2, respectively. The cooperativity of the sodium dependence allows the terreactant model to be reduced to a bireactant model in which the sodium concentration only appears mathematically as the square of the sodium concentration. The data then fit the relationship

The action of Nupercaine on calcium efflux from rat liver mitochondria.

1. Nupercaine inhibits the Ca2+ efflux from rat liver mitochondria observed in the presence of Ruthenium Red, 50% inhibition being obtained at 80 microM-Nupercaine. 2. Neither the Ruthenium Red-stimulated efflux nor its inhibition by Nupercaine can be directly attributed to effects on mitochondrial stability. 3. Nupercaine perturbs the steady-state external Ca2+ concentration in the absence of ...

The inhibition of mitochondrial calcium transport by lanthanides and ruthenium red.

An EGTA (ethanedioxybis(ethylamine)tetra-acetic acid)-quench technique was developed for measuring initial rates of (45)Ca(2+) transport by rat liver mitochondria. This method was used in conjunction with studies of Ca(2+)-stimulated respiration to examine the mechanisms of inhibition of Ca(2+) transport by the lanthanides and Ruthenium Red. Ruthenium Red inhibits Ca(2+) transport non-competiti...

Estradiol affect Na-dependent Ca2+ efflux from synaptosomal mitochondria.

The effects of gonadal steroid hormone, 17beta-estradiol (E2), in vitro on rat brain mitochondria Ca2+ movement were investigated. Intrasynaptosomal mitochondria Ca2+ uptake via an energy-driven Ca2+ uniporter have Km = 112.73 +/- 7.3 micromol x l(-1) and Vmax = 21.97 +/- 1.7 nmol 45Ca2+ mg(-1). Ca2+ release trough a Na+/Ca2+ antiporter was measured with a Km for Na+ of 43.7 +/- 2.6 mmol x l(-1...

In vitro cellular muscle calcium metabolism. Characterization of effects of 1,25-dihydroxy-vitamin D3 and 25-hydroxy-vitamin D3.

Cultures of vitamin D-deficient chick soleus muscle and 12 day-old chick embryo myoblasts were used to characterize the effects of 1,25-dihydroxy-vitamin D3 and 25-hydroxy-vitamin D3 on muscle cell Ca metabolism. Physiological amounts of both sterols increased the rate and extent of 45Ca uptake by cultures. However, 1,25(OH)2D3 was significantly more effective than 25 OHD3. The greater potency ...

Calcium binding and translocation by the voltage-dependent anion channel: a possible regulatory mechanism in mitochondrial function.

Mitochondria play a central role in energy metabolism, Ca(2+) signalling, aging and cell death. To control cytosolic or mitochondrial Ca(2+) concentration, mitochondria possess several Ca(2+)-transport systems across the inner membrane. However, the pathway for Ca(2+) crossing the outer membrane has not been directly addressed. We report that purified voltage-dependent anion channel (VDAC) reco...
